- Crude peptide / Desalted peptide
- Immunograde peptide (purity >75%): the crude peptide is precipitated and washed several times for the complete extraction of organic impurities and to remove most secondary products. This level of purity of the peptides is usually enough for generating or testing antibodies.
- Purified peptides (purity >85%): further purification by preparative HPLC allows the elimination of peptides resulting from incomplete coupling (i.e. peptide lacking one or more amino acids). The 85% purified peptides can be used for antiserum production, ELISA antibody titration and monoclonal antibody production for example.
- Highly purified peptide (purity >95%): this level of purity is required for most biological or enzyme activity studies.
- Purity > 98% or > 99% can also be performed.

ProteoGenix’ fmoc solid phase peptide synthesis technology
ProteoGenix mainly uses fmoc solid phase peptide synthesis technology to produce the required amino acid sequences. It consists of assembling every fmoc protected amino acid via a specific resin. Once the molecule is assembled, it is first cleaved from the resin and then deprotected from the fmoc group also known as fluorenylmethoxycarbonyl group. The obtained crude product can then be purified by HPLC to get the desired purity from 70% to 99%. Other protection groups can also be used like t-Boc but fmoc leaves much less impurities in the final product and allows to get higher yields. It is therefore the best current method to reach our high quality production standards. This technology is widely used and has proven for the last 30 years to be the most reliable method available on the market.
We use many coupling reagents like BOP reagent, HBTU, HOBt, TBTU. Our most commonly used protecting reagents are Fmoc-Cl, Fmoc-OSu. We use linkers for solid phase method such as HMP linker, DHP linker, Rink amide linker.
In our manufactures, our scientists are using both manual and automated techniques but the flexibility of manual methods allows us to be able to deliver longer sequences.

Multiple Antigenic Peptides or MAP are commonly used for generating monoclonal or polyclonal antibodies. It is an alternative antigen type to single peptides, native proteins, recombinant proteins or even cDNA injections. Each molecule is bound to a matrix composed by 4 or 8 lysine residues branches. One of the main advantages is that it doesn’t require any further conjugation to a BSA or KLH carrier.
A peptide library is usually composed by chemical sequences of 15 to 20 AA that cover a whole protein no matter the size. Given our high production capacity there is no limitation in protein size. One of the most popular applications is to perform antibody epitope mapping to determine precisely where an antibody binds to a protein. But one should keep in mind that it only makes sense for linear epitopes and not for conformational epitopes. It is very important to define the right parameters such as the number of overlapping amino acids for each sequence and the size of the final sequences. MAPs are also widely used for drug discovery and especially during drug target screening steps.
