TNF-alpha, also known as tumor necrosis factor-alpha, is a cytokine protein produced mainly by macrophages and other white blood cells. It is involved in the regulation of immune responses, inflammation, and cell death. TNF-alpha binds to two receptors, TNFR1 and TNFR2, which are present on the surface of many cell types. Binding to TNFR1 activates the pro-inflammatory signals, while binding to TNFR2 leads to cell death.
TNF-alpha is a target for antibody-drugs used to treat autoimmune diseases such as rheumatoid arthritis, psoriasis, and Crohn’s disease. These drugs are monoclonal antibodies that bind to TNF-alpha and inhibit its activity, thus reducing inflammation and preventing tissue damage. Infliximab and adalimumab are two of the most commonly used TNF-alpha inhibitors.
TNF-alpha has also been studied for its potential therapeutic applications. It has been used in cancer therapy to induce apoptosis in tumor cells, and it has been studied as a potential treatment for HIV infection.
