Thioredoxin reductase 1 (cytoplasm) is an enzyme in humans encoded by the TXNRD1 gene, which encodes a member of the pyridine nucleotide oxidoreductase family. The protein can reduce thioredoxin and other substrates, and play a role in selenium metabolism and anti-oxidative stress. This functional enzyme is considered to be a homodimer using FAD as a cofactor. Each subunit contains selenocysteine (Sec), which is necessary for catalytic activity. Selenocysteine is encoded by the UGA codon which normally signals the termination of translation. The 3′ UTR of selenocysteine-containing genes have a common stem-loop structure, the sec insertion sequence (SECIS), that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. Alternative splicing results in several transcript variants encoding the same or different isoforms.
