How are antibodies developed?

Polyclonal Antibody Development: In this approach, animals are immunized with the antigen causing the immune system of the animal to respond by producing a diverse pool of antibodies targeting various epitopes on the antigen. Blood is collected from the immunized animal, and the serum containing a mixture of antibodies is isolated.

Polyclonal antibodies provide broad recognition of the antigen, making them suitable for various applications in research and diagnostics.

Monoclonal Antibody Development: This process aims to produce highly specific antibodies that target a single epitope (a specific region) on a particular antigen. Unlike polyclonal antibodies which are a mixture of different antibodies targeting various epitopes, monoclonal antibodies are derived from a single B cell clone, resulting in a homogenous population of antibodies with identical binding properties.

  • Antibody Phage Display: A library of antibody genes is fused to the gene sequence of a bacteriophage coat protein causing the antibody to be displayed on the surface of bacteriophages. The phage library is then screened for binding to the target antigen in vitro. The phages displaying the desired antibodies are selected, and the genes encoding these antibodies are isolated for further production (antibody genes are inside the bacteriophage). Phage display offers the advantage of identifying antibodies with high affinity and specificity for the target antigen.
  • Single B Cell Isolation: This technique involves isolating individual B cells from immunized animals or individuals based on their ability to produce antibodies against the target antigen. These selected B cells are then used to generate monoclonal antibodies, allowing researchers to specifically target B cells that produce the desired antibodies.
  • Hybridoma Cell Line Generation: This classical method involves immunizing an animal with the target antigen to elicit an immune response. B cells producing the desired antibodies are then isolated from the immunized animal. These B cells are fused with immortal myeloma cells to create hybridoma cells. The hybridomas can continuously produce the desired monoclonal antibodies with high specificity and reproducibility. Hybridoma technology is well-established and has been widely used for the development of monoclonal antibodies.
