    Western Blot

    Western blotting (also known as protein immunoblotting) is a widely used technique for the detection and characterization of proteins. The technique involves the separation of proteins by size through electrophoresis, followed by transfer of the proteins to a membrane, and finally detection of the proteins with antibodies specific to the target protein of interest.

    Protein Separation

    The first step in a Western blot is to separate the proteins of interest by size through electrophoresis. This is done by placing a sample containing the proteins of interest onto a gel and then applying an electric current to the gel. The proteins are then separated according to their size, with smaller proteins migrating faster than larger proteins.

    Membrane Transfer

    Once the proteins have been separated by size, they are transferred from the gel to a membrane. This is usually done by sandwiching the gel between the membrane and a stack of filter papers, and then applying a current to the membrane. This causes the proteins to be transferred from the gel onto the membrane.

    Antibody Detection

    The final step in a Western blot is the detection of the proteins of interest with antibodies specific to the target protein. To do this, the membrane is first incubated with a primary antibody that binds to the target protein. This is followed by the addition of a secondary antibody that is conjugated to a reporter molecule, such as an enzyme or fluorescent dye. This allows the proteins of interest to be visualized.
