XtenCHO ™ Transfection kit
Transfection is the key step of recombinant proteins production in eukaryotic cells. The aim of this technique is to inject nucleic acid strains into the cells. The coding material thus implemented will use the cell own machinery to express its proteins. This step is highly costing and dangerous to the cells. Therefore, different technologies have been developed to enable the researchers to achieve it succefully. Proteogenix has engineered its own proprietary cell line XtenCHO™ to use it for its recombinant productions. It is now available to its customers. The XtenCHO™ Transfection Kit has been specifically optimized to support the transient transfection of XtenCHO™ Cells. Thanks to this tool the user can benefit from the unprecedented high yields of this cell line.
The XtenCHO™ Transfection Kit is composed of XtenFect Reagent and the XtenCHO™ Enhancer. The XtenFect Reagent is optimized for the transfection of XtenCHO™ cell cultures grown in XtenCHO™ Expression Medium. It enables the Chinese Hamster Ovarian (CHO) cells to efficiently take on the nucleic acids. This step is critical to reach the highest recombinant protein production yields with XtenCHO™ Expression System. The XtenCHO™ Enhancer is optimized to maintain high viability in high density transfected XtenCHO™ Cells. This leads to extended production time and higher yields per liter of culture.
The XtenFect Reagent is provided as a 10 times concentrated stock solution that has to be diluted in the provided XtenFect Working Solution. After being reconstituted, the XtenFect working solution can be directly added to the XtenCHO™ cell culture. On the contrary to other transfection reagents, it does not need a previous step of complexe formation with DNA, making the process easier and faster. This product is intended for 1L of culture. These products are animal component free and chemically composed.
The XtenCHO™ Transfection Kit is a component of the XtenCHO™ Starter Kit.
Therapeutic antibodies from several IgG subclasses and isotypes (IgG1-kappa and lambda, IgG2-lambda, IgG4-kappa) were produced with XtenCHO ™ cells and in parallel with CHO-S ™ and ExpiCHO ™ expression systems. XtenCHO ™ cells were transfected with ProteoGenix' proprietary TGE system (transfection protocol, transfection reagent and culture medium). CHO-S ™and ExpiCHO ™ cells were transfected according to manufacturer's instructions for each expression system (protocol, reagents and culture medium). Determination of production yields was done with an Octet Red96 system (Protein G biosensor).
