TNF-alpha: Structure, Activity, and Application

TNF-alpha (tumor necrosis factor-alpha) is a cytokine protein produced by various cells in the body. It is a type of pro-inflammatory cytokine, meaning it plays an important role in the body’s immune response.

Structure

TNF-alpha is a homotrimeric glycoprotein composed of three identical subunits, each around 17 kDa in size. Each subunit is composed of an N-terminal signal peptide, a pro-region, and a mature region.

Activity

TNF-alpha is involved in a variety of cellular processes, including cell differentiation, apoptosis, and the regulation of immune responses. It binds to two distinct receptors, TNFR1 and TNFR2, and activates a variety of intracellular signaling pathways.

Application

TNF-alpha is a key mediator of inflammation, and has been implicated in a variety of diseases, including rheumatoid arthritis, psoriasis, and Crohn’s disease. As such, it has become a major target for therapeutic intervention. A number of TNF-alpha inhibitors have been developed for the treatment of these conditions.
