Phage display? What is this?


This technology includes determination of interaction partners of a protein, which would be used as the immobilized phage "bait" with a DNA library (consisting of all coding sequences of an organism) so that the function of that protein may be determined. As such, phage display is a useful tool in drug discovery by using libraries composed of millions of antibody expressed on phage, with a view to isolate antibodies highly specific against an antigen.


Advantages of phage display

The main advantage of phage display is the possibility to screen a library encompassing a huge pool of antibody variants that are all potential binders. As we consider that only 10% of the antigens are immunogenic, this technology offers a great alternative to develop new antibodies for biotherapies but also diagnostics or even only fundamental research.

The phage display offers several advantages compared to the standard “hybridoma” method :

  • It’s not necessary to start the antibodies development with animals immunizations.
  • →We are involved in less animals use for antibodies development. Alternative methods like Phage Display exist and must be emphasized.

  • Larger host species choice for your antibodies development!
  • →The antibodies libraries can be obtained from many species, and enables you to develop antibodies from non-standard laboratory hosts (camel, sheep, alpaca), or from human.

  • NO FUSION and NO LIMITING DILUTIONS.
  • →Everyone who already developed antibodies knows fusion and limiting dilutions are the downsides of the hybridoma method. Without such steps, there is no risk to lose a good candidate along the way.

  • Humanized antibodies. The key to success for potential therapeutic mAbs.


Once best binders are identified, they are subcloned, expressed and sequenced for further characterization in different applications potentially followed by affinity maturation. We are producing highly specific and high affinity antibodies from various species including camel, alpaca, rabbit, mouse, sheep and human. Depending on your needs, we propose to use naïve or immune libraries with up to 1010 available clones.

For bio therapeutic antibodies, we therefore don’t need to perform any humanization as the library that we screen can be already human. However, we usually engineer the sequence to increase the production yield of the antibody, optimize the stability of the molecule and increase its affinity.
